Clinical evidence comes mainly from small studies conducted in Russia, several of which were open-label or lacked robust blinding. Reported outcomes include lower anxiety scores, changes in attention measures, and effects on asthenic states following illness. Sample sizes are typically in the tens of participants, and independent replication outside the region is scarce. Reviews published in English generally note the limited methodological quality of the underlying trials. Whether the compound produces clinically meaningful effects under rigorous conditions remains unresolved.
Proposed mechanisms center on modulation of the GABA system, but no single molecular target has been confirmed. Rodent studies report changes in GABA-A receptor expression and in the turnover of serotonin, dopamine, and norepinephrine in several brain regions. Increases in brain-derived neurotrophic factor and its receptor have also been described after repeated administration. These findings come largely from animal models, and the degree to which they describe human neurochemistry remains an open question. The mechanism is best characterized as multi-system and not fully resolved.
Solubility behavior is a practical concern for handling. Selank dissolves readily in water and in common aqueous buffers, which simplifies preparation of working solutions. The choice of solvent, ionic strength, and pH can influence aggregation over time, particularly at higher concentrations. Aqueous solutions are typically sterile-filtered before use. Because stability depends on several variables, storage and handling notes should be treated as general guidance rather than fixed rules, and specific values are best confirmed against a certificate of analysis for each batch.
Characterization of Selank in a laboratory setting relies on standard peptide methods. Reverse-phase high-performance liquid chromatography separates the target from related impurities and provides a purity figure, commonly reported as 95 percent or higher. Mass spectrometry, typically electrospray ionization or matrix-assisted laser desorption, confirms the molecular mass and helps detect truncation or modification. Amino acid analysis can verify composition when a sequence-level check is needed. These techniques together establish identity and purity for a given lot.

Published clinical work is concentrated in Russian-language journals and generally involves small samples without independent replication. Systematic reviews in English note the shortage of randomised, placebo-controlled trials and the difficulty of verifying methods from translated reports. Outcome measures vary between studies, which complicates pooling of results. Interest in the compound as a cognitive or anxiolytic agent therefore rests on a thinner evidence base than the volume of citations suggests. Replication in well-powered trials with preregistered endpoints would be needed before firm conclusions about efficacy can be drawn.
Proposed mechanisms centre on the GABAergic system. Animal and tissue studies report changes in GABA-A receptor expression and reduced activity of GABA transaminase, the enzyme that degrades GABA. Effects on monoamine turnover, including serotonin and dopamine pathways, are also described, and a separate line of work links the peptide to increased expression of brain-derived neurotrophic factor in hippocampal tissue. Most of these findings come from rodent models and cell preparations. How the individual observations combine into a single coherent mode of action is not settled.
Pharmacokinetic data are sparse and largely derived from animal work. After intranasal administration the peptide appears in plasma within minutes, and reported half-lives are short, on the order of minutes to tens of minutes. Degradation proceeds through ordinary proteolytic cleavage into constituent amino acids and smaller fragments. Direct evidence that intact Selank reaches brain tissue in meaningful amounts is limited, and the extent of blood-brain barrier penetration is debated. Some authors argue that fragments, not the parent peptide, carry much of the observed activity.
Regulatory status differs sharply by region. Selank holds a Russian marketing authorization, where it is supplied mainly as nasal drops, while authorities elsewhere have not approved it for medical use. Material sold internationally is therefore usually labeled as a research chemical rather than a medicine. Peer-reviewed publications come predominantly from Russian laboratories, and sample sizes are generally small. Whether the compound produces comparable effects under independent, well-controlled replication remains an open question that the broader literature has not settled.
Selank is a synthetic heptapeptide developed in Russia as a structural analogue of tuftsin, a naturally occurring immunomodulatory tetrapeptide. Its sequence, Thr-Lys-Pro-Arg-Pro-Gly-Pro, keeps the tuftsin core at the N-terminus and appends a Pro-Gly-Pro tail. Researchers at the Institute of Molecular Genetics in Moscow synthesized the compound during the 1990s while searching for peptides with combined anxiolytic and immunomodulatory activity. The added tail was intended to resist enzymatic cleavage and prolong the molecule's presence in circulation.
The compound has a calculated molecular weight near 751.9 daltons and carries a net positive charge at physiological pH because of its arginine residue. It dissolves freely in water and in common aqueous buffers, and typically appears as a white or off-white lyophilized powder. The amide backbone makes the molecule susceptible to peptidases, which limits oral use and favors intranasal or parenteral routes. Nomenclature in the literature varies: the substance is also described by the sequence abbreviation TP-7 and by a Russian trade designation.
Handling follows standard practice for research peptides. Material is weighed in a low-humidity environment because the powder absorbs atmospheric moisture. Purity is reported as the percentage area of the main peak in a chromatogram, with specifications commonly set at 95 percent or higher; values below that threshold indicate the presence of truncated or modified species. Residual trifluoroacetate from purification is often present and may affect mass balance. Certificates of analysis should state the analytical method, the column and gradient used, and the lot-specific retention time so that results can be compared across suppliers.
Identity and purity of selank are established with reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The peptide elutes from C18 columns with acetonitrile gradients in water containing trifluoroacetic acid or formic acid, and detection is usually performed by ultraviolet absorbance near 214 nm. Electrospray ionization in positive mode gives a doubly protonated ion near m/z 377, consistent with a mass of about 752 Da. Amino acid analysis or tandem mass spectrometry of fragment ions confirms the sequence. Because the molecule has no aromatic residues, it lacks a usable 280 nm chromophore, so low-wavelength detection or mass spectrometry is required.
Peptide bonds in selank are susceptible to hydrolysis under strongly acidic or basic conditions, and the terminal proline residues are vulnerable to exopeptidase activity in biological samples. Lyophilized powder stored dry at -20 °C typically remains stable for extended periods, whereas aqueous solutions degrade faster and may lose measurable purity within days to weeks depending on pH, temperature, and microbial load. Repeated freeze-thaw cycles promote aggregation and adsorption to container surfaces. For analytical work, solutions are usually prepared fresh, kept cold, and used within a single working day.
Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres is the standard purity method. Mass spectrometry, typically electrospray ionisation, confirms identity through the expected mass-to-charge pattern. Amino acid analysis can verify composition independently. Chiral purity requires separate techniques such as derivatisation followed by chromatographic separation, and such data are rarely reported for research-grade material.
Quantification in biological matrices relies on liquid chromatography coupled to tandem mass spectrometry with stable-isotope internal standards. Low plasma concentrations and adsorption to container surfaces both complicate measurement. Solid-phase extraction is often needed to reduce matrix interference before injection. Reported limits of quantification differ widely between laboratories, which makes direct comparison of pharmacokinetic results difficult and limits meta-analysis.
Peptide bonds are vulnerable to protease attack, and Selank is no exception. Measured half-life in serum is short, on the order of minutes in several reports, which explains why intranasal administration is the common route described in the literature. Absorption across the nasal mucosa partially bypasses first-pass hepatic metabolism. Quantitative data on human bioavailability remain limited and are difficult to compare across studies.
Skeletons excavated from the Coburn Street Burial Ground (1750 to 1827 CE) in Cape Town, South Africa, were analyzed using stable isotope data in order to determine geographical histories and life histories. The people buried in this cemetery were assumed to be slaves and members of the underclass based on the informal nature of the cemetery; biomechanical stress analysis and stable isotope analysis, combined with other archaeological data, seem to support this supposition. Based on stable isotope levels, one study reported that eight Cobern Street Burial Ground individuals consumed a diet based on C4 (tropical) plants in childhood, then consumed more C3 plants, which were more common there later in their lives. Six of these individuals had dental modifications similar to those carried out by peoples inhabiting tropical areas known to be targeted by slavers who brought enslaved individuals from other parts of Africa to the colony. Based on this evidence, it was argued that these individuals represent enslaved persons from areas of Africa where C4 plants were consumed and who were brought to the Cape as laborers. These individuals were not assigned to a specific ethnicity, but similar dental modifications are carried out by the Makua, Yao, and Marav peoples. Four individuals were buried with no grave goods, in accordance with Muslim tradition, facing Signal Hill, which is a point of significance for local Muslims.
was renamed Merck Sharp & Dohme, and Schering-Plough renamed as "Merck & Co., Inc." The maneuver was an attempt to avoid a "change-of-control" in order to preserve Schering-Plough's rights to market Remicade. A settlement with Johnson & Johnson was reached in 2011, in which Merck agreed to pay $500 million. Merck Sharp & Dohme remains a subsidiary of the Merck & Co. parent. Richard Clark retired as CEO and company president in October 2011 and Kenneth Frazier became CEO. In October 2013, Merck announced it would cut 8,500 jobs in an attempt to cut $2.5 billion from its costs by 2015. Combined with 7,500 job cuts announced in 2011 and 2012, the layoffs amounted to 20% of its workforce. By 2014, research performed at Merck has led to U.S. FDA approval of 63 new molecular entities. In August 2014, Merck acquired Idenix Pharmaceuticals for $3.85 billion. In September 2014, the US Food and Drug Administration (FDA) approved Pembrolizumab (MK-3475) as a breakthrough therapy for melanoma treatment. In clinical trials, pembrolizumab provided partial tumor regression in about one quarter of patients, many of whom have not seen further progression of their disease in over 6 months of follow-up. In December 2014, the company acquired Swiss biotechnology company OncoEthix for up to $375 million. Between 2010 and 2015, the company cut around 36,450 jobs. During that time, the company sold its consumer health business to Bayer and narrowed the company's focus to immunology, vaccines, diabetes, emerging markets and medicines used in hospitals, like certain antibiotics.
=== Stimulus-secretion coupling === In response to, for instance, a rise in the plasma sodium concentration, vasopressin neurons also discharge action potentials in bursts, but these bursts are much longer and are less intense than the bursts displayed by oxytocin neurons, and the bursts in vasopressin cells are not synchronised. It seemed strange that the vasopressin cells should fire in bursts. As the activity of the vasopressin cells is not synchronised, the overall level of vasopressin secretion into the blood is continuous, not pulsatile. Richard Dyball and his co-workers speculated that this pattern of activity, called "phasic firing", might be particularly effective for causing vasopressin secretion. They showed this to be the case by studying vasopressin secretion from the isolated posterior pituitary gland in vitro. They found that vasopressin secretion could be evoked by electrical stimulus pulses applied to the gland, and that much more hormone was released by a phasic pattern of stimulation than by a continuous pattern of stimulation. These experiments led to interest in "stimulus-secretion coupling" - the relationship between electrical activity and secretion. Supraoptic neurons are unusual because of the large amounts of peptide that they secrete, and because they secrete the peptides into the blood. However, many neurons in the brain, and especially in the hypothalamus, synthesize peptides. It is now thought that bursts of electrical activity might be generally important for releasing large amounts of peptide from peptide-secreting neurons.
WOSM's bureau is its secretariat, which carries out the instructions of its conference and committee. The bureau is administered by the secretary general, supported by a staff of technical resource personnel. A bureau was established in London, England in 1922, moved to Ottawa, Ontario, Canada in 1959, Geneva, Switzerland after 1 May 1968 and Kuala Lumpur after August 2013.
Recently, copper alloys have become important netting materials in aquaculture because they are antimicrobial (i.e., they destroy bacteria, viruses, fungi, algae, and other microbes) and they therefore prevent biofouling (i.e., the undesirable accumulation, adhesion, and growth of microorganisms, plants, algae, tubeworms, barnacles, mollusks, and other organisms). By inhibiting microbial growth, copper alloy aquaculture cages avoid costly net changes that are necessary with other materials. The resistance of organism growth on copper alloy nets also provides a cleaner and healthier environment for farmed fish to grow and thrive.

In April 1805, Britain and Russia signed a treaty with the aim of removing the French from the Batavian Republic (roughly present-day Netherlands) and the Swiss Confederation. Austria joined the alliance after the annexation of Genoa (Ligurian Republic) and the proclamation of Napoleon as King of Italy on 17 March 1805. Sweden, which had already agreed to lease Swedish Pomerania as a military base for British troops against France, entered the coalition on 9 August. The Austrians began the war by invading Bavaria on 8 September 1805 with an army of about 72,000 under Karl Mack von Leiberich, and the French army marched out from Boulogne in late July 1805 to confront them. At Ulm (25 September – 20 October) Napoleon surrounded Mack's army, forcing its surrender without significant losses. With the main Austrian army north of the Alps defeated (another army under Archduke Charles fought against André Masséna's French army in Italy), Napoleon occupied Vienna on 13 November. Far from his supply lines, he faced a larger Austro–Russian army under the command of Mikhail Kutuzov, with Emperor Alexander I of Russia personally present. On 2 December, Napoleon crushed the Austro–Russian force in Moravia at Austerlitz (usually considered his greatest victory). He inflicted 25,000 casualties on a numerically superior enemy army while sustaining fewer than 7,000 in his own force.

=== Transthyretin-related hereditary amyloidosis === The FDA granted Patisiran (Onpattro) orphan drug status and breakthrough therapy designation due to its novel mechanism involving RNA therapy to block the production of an abnormal form of transthyretin. Patisiran received full FDA approval in 2018 and its RNA lipid nanoparticle drug delivery system was later used in the Pfizer–BioNTech COVID-19 vaccine and Moderna RNA vaccines.
=== Intravenous administration === Serum vancomycin levels may be monitored in an effort to reduce side effects, but the value of such monitoring has been questioned. Peak and trough levels are usually monitored, and for research purposes the area under the concentration curve is also sometimes used. Toxicity is best monitored by looking at trough values. Immunoassays are commonly used to measure vancomycin levels. Common adverse drug reactions (≥1% of patients) associated with intravenous vancomycin include:
== Side effects == In September 2020, the US Food and Drug Administration (FDA) required the boxed warning be updated for all benzodiazepine medicines to describe the risks of abuse, misuse, addiction, physical dependence, and withdrawal reactions consistently across all the medicines in the class.

=== Food === Protein hydrolysis release savory free amino acids (especially glutamic acid) and peptides. Hydrolyzed vegetable protein and yeast extract are commonly used as flavor enhancers (sources of umami) as a result. The non-protein components in these products also contribute to the flavor. Protein hydrolysis also increases their digestibility and rate of digestion. Some hydrolyzed beef protein powders are used for specialized diets for athletes. Protein hydrolysis can be used to destroy epitopes involved in recognition by antibodies involved in allergy. "An allergen must have at least 2 IgE-binding epitopes, and each epitope must be at least 15 amino acid residues long, to trigger a type 1 hypersensitivity reaction." As a result, it has been used to reduce the allergenicity of infant formula: Reducing the size of cow milk proteins in the formula makes it more suitable for consumption by babies suffering from milk protein intolerance. The US FDA has approved a label for this usage of partially-hydrolyzed proteins in 2017, but a meta-analysis published the same year shows insufficient evidence for this use.
=== Economic and social cost === Addiction imposes substantial financial and human costs on individuals and on society. These costs arise from the direct adverse effects of drugs and associated healthcare costs (e.g., emergency medical services and outpatient and inpatient care), long-term complications (e.g., lung cancer from smoking tobacco products, liver cirrhosis and dementia from chronic alcohol consumption, and meth mouth from methamphetamine use), the loss of productivity and associated welfare costs, fatal and non-fatal accidents (e.g., traffic collisions), suicides, homicides, and incarceration, among others. In the United States, an estimated 69,973 people died of drug overdose in 2025, down almost 14% from 81,313 in 2024 and the third consecutive annual decline following the peak reached during the opioid epidemic. Access to treatment is limited worldwide: among countries reporting data to the World Health Organization, the proportion of people with a substance use disorder in contact with treatment services ranged from under 1% to no more than 35%.
Trazodone may act predominantly as a 5-HT2A receptor antagonist to mediate its therapeutic benefits against anxiety and depression. Its inhibitory effects on serotonin reuptake and 5-HT2C receptors are comparatively weak. In relation to these properties, trazodone does not have similar properties to selective serotonin reuptake inhibitors (SSRIs) and is not particularly associated with increased appetite and weight gain – unlike other 5-HT2C antagonists like mirtazapine. Moderate 5-HT1A partial agonism may contribute to trazodone's antidepressant and anxiolytic actions to some extent as well. The combined actions of 5-HT2A and 5HT2C receptor antagonism with serotonin reuptake inhibition only occur at moderate to high doses of trazodone. Doses of trazodone lower than those effective for antidepressant action are frequently used for the effective treatment of insomnia. Low doses exploit trazodone's potent actions as a 5-HT2A receptor antagonist, and its properties as an antagonist of H1 and α1-adrenergic receptors, but do not adequately exploit its SERT or 5-HT2C inhibition properties, which are weaker. Since insomnia is one of the most frequent residual symptoms of depression after treatment with an SSRI, a hypnotic is often necessary for patients with a major depressive episode. Not only can a hypnotic potentially relieve the insomnia itself, but treating insomnia in patients with major depression may also increase remission rates due to the improvement of other symptoms such as loss of energy and depressed mood.
==== Cardiovascular ==== Other side-effects can include alterations in the structure of the heart, such as enlargement and thickening of the left ventricle, which impairs its contraction and relaxation, and therefore reducing ejected blood volume. Possible effects of these alterations in the heart are hypertension, cardiac arrhythmias, congestive heart failure, heart attacks, and sudden cardiac death. These changes are also seen in non-drug-using athletes, but steroid use may accelerate this process. However, both the connection between changes in the structure of the left ventricle and decreased cardiac function, as well as the connection to steroid use have been disputed. AAS use can cause harmful changes in cholesterol levels: Some steroids cause an increase in LDL cholesterol and a decrease in HDL cholesterol.

Reports describe effects on GABA-A receptor expression, monoamine turnover, and neurotrophic factor levels. These are proposed mechanisms drawn mainly from animal models. No single molecular target has been established.
Intranasal delivery is the route described in most published work. It is used because the peptide is degraded quickly once it reaches circulation. Direct evidence of brain penetration in humans is lacking.
Most trials are small, regionally concentrated, and often lack rigorous blinding or placebo control. Independent replication is limited. Reviews in English generally rate the evidence as preliminary.
Reverse-phase HPLC is the usual method and gives a percentage purity value. Mass spectrometry then confirms the molecular mass. Together they provide a basic identity and purity profile for a lot.
