Adding a proline-rich tail to the short parent peptide was a deliberate strategy, because proline residues restrict the conformations available to many peptidases. Peptide bonds in selank are susceptible to hydrolysis under strongly acidic or basic conditions, and the terminal proline residues are vulnerable to exopeptidase activity in biological samples. Lyophilized powder stored dry at -20 °C typically remains stable for extended periods, whereas aqueous solutions degrade faster and may lose measurable purity within days to weeks depending on pH, temperature, and microbial load. The plantar fascia contributes to support of the arch of the foot by acting as a tie-rod, where it undergoes tension when the foot bears weight. One biomechanical model estimated it carries as much as 14% of the total load of the foot. In an experiment using cadavers, it was found that failure of the plantar fascia averaged at loads of 118±244 newtons (12 ± 25 kilograms-force; 27 ± 55 pounds-force). leucine zipper (ZIP) A common structural motif in DNA-binding transcription factors and some other types of proteins, approximately 35 amino acids in length, characterized chiefly by the recurrence of the amino acid leucine every seven residues. When modeled in an idealized alpha-helical conformation, the leucine residues are positioned in such a way that they can interdigitate with the same or similar motifs in an alpha helix belonging to another similar polypeptide, facilitating dimerization and the formation of a complex resembling a zipper.
Sanger's reagent proved effective at labelling the N-terminal amino group at one end of the polypeptide chain. He then partially hydrolysed the insulin into short peptides, either with hydrochloric acid or using an enzyme such as trypsin. The mixture of peptides was fractionated in two dimensions on a sheet of filter paper, first by electrophoresis in one dimension and then, perpendicular to that, by chromatography in the other. The different peptide fragments of insulin, detected with ninhydrin, moved to different positions on the paper, creating a distinct pattern that Sanger called "fingerprints". The peptide from the N-terminus could be recognised by the yellow colour imparted by the FDNB label and the identity of the labelled amino acid at the end of the peptide determined by complete acid hydrolysis and discovering which dinitrophenyl-amino acid was there. By repeating this type of procedure Sanger was able to determine the sequences of the many peptides generated using different methods for the initial partial hydrolysis. These could then be assembled into the longer sequences to deduce the complete structure of insulin. Finally, because the A and B chains are physiologically inactive without the three linking disulfide bonds (two interchain, one intrachain on A), Sanger and coworkers determined their assignments in 1955. Sanger's principal conclusion was that the two polypeptide chains of the protein insulin had precise amino acid sequences and, by extension, that every protein had a unique sequence.

A great deal of attention has focused, understandably, on use by individuals with diabetes and their relatives, as well as by health-care professionals such as diabetologists / endocrinologists and diabetes educators. However, an important group of health-carers involved in the provision of day-today care for many people with diabetes are primary care physicians (general practitioners [GPs]). A workshop was held in September 2000 in Italy — by an independent diabetologist / endocrinologist unconnected with the AIDA simulator's development — to gain experience with application of the AIDA diabetes simulation approach as a teaching tool for general practitioners (GPs). Feedback obtained from participants attending the workshop was very positive, with GPs reporting the simulation approach to be both of interest and use.
